Cambridge Principal Examiner Reports for Biology 0610 identify the same weaknesses year after year. Understanding these helps you avoid losing marks.

Cell biology and microscopy

Confusing structures in photomicrographs

Candidates often misidentify cell structures, especially confusing mitochondria with ribosomes. Both appear as small organelles but have different functions.

Fix: Mitochondria = site of aerobic respiration; elongated. Ribosomes = site of protein synthesis; smaller, in cytoplasm or on RER.

Osmosis experiment errors

Many incorrectly assume solutions in test tubes are water when a solute is present. They fail to recognise water potential and the direction of water movement.

Fix: Always identify the solute. Water moves from higher to lower water potential (into more concentrated solutions).

Magnification calculations

Candidates multiply when they should divide (or vice versa) for actual size = image size ÷ magnification.

Fix: Learn the formula. Check units (mm, µm, nm). Show working.

Biochemistry and digestion

Reducing sugar tests

Uncertainty about positive results (brick-red precipitate with Benedict’s) and conditions (heat, reducing sugars vs. non-reducing).

Fix: Benedict’s + reducing sugar + heat → brick-red. Sucrose is non-reducing unless hydrolysed first.

Amylase and starch digestion

Confusion about products: starch breaks down into maltose (disaccharide), not directly into glucose.

Fix: Starch → maltose (amylase); maltose → glucose (maltase).

Enzyme pH

Candidates forget that stomach enzymes work at low pH; intestinal enzymes at higher pH. Optimal pH is enzyme-specific.

Fix: Pepsin → low pH; pancreatic amylase, trypsin → neutral/alkaline.

Transport and diffusion

Concentration gradient

Incorrect belief that decreasing concentration gradient increases diffusion rate. Higher gradient = faster diffusion.

Fix: Diffusion rate ∝ concentration gradient. Steeper = faster.

Active transport

Some say active transport occurs without a membrane. It must occur across a membrane, against the gradient, using energy.

Fix: Active transport: across membrane; against gradient; requires ATP.

Human systems

Kidney vs. bladder

Confusion about urea excretion: kidneys filter and produce urine; bladder stores it. Urea is excreted by the kidneys.

Fix: Kidney = excretion (urea in urine). Bladder = storage.

Menstrual cycle

Uncertainty about uterine lining changes. Before ovulation, lining thickens (builds up); it breaks down if no implantation.

Fix: Follicular phase: lining builds. Luteal: maintains. Menstruation: sheds if no pregnancy.

Plant biology

Seed germination

Many think sunlight is required. Germination needs water, oxygen and suitable temperature—not light (initially).

Fix: Water, oxygen, warmth. Light needed later for photosynthesis.

Transpiration

Uncertainty about where evaporation occurs: from spongy mesophyll cell surfaces into air spaces.

Fix: Water evaporates from mesophyll into air spaces; diffuses out through stomata.

Classification and genetics

Arthropod classification

Misidentifying arthropods—e.g. classifying arachnids (spiders) as crustaceans. Arachnids have 8 legs; crustaceans often have more, cephalothorax + abdomen.

Fix: Arachnids: 8 legs, 2 body parts. Crustaceans: many legs, often aquatic. Insects: 6 legs, 3 body parts.

Environmental vs. genetic variation

Height variation is affected by both genes and environment (nutrition, health). Not only genetic.

Fix: Phenotype = genotype + environment.

General exam technique

  • Read questions carefully—work through experimental scenarios step by step
  • Examine diagrams and graphs before answering
  • State = short; Describe = what/how; Explain = why
  • Suggest = apply to the scenario—generic answers score poorly
